Nuclear speckles and paraspeckles are ribonucleoprotein complexes built on long non-coding RNAs (lncRNAs) that undergo phase separation. These nuclear condensates activate the transcription and splicing of a subset of stress-related genes. To regulate this response, we hypothesize that nuclear speckles and paraspeckles respond to selective stress conditions by recruiting and scaffolding DNA and RNA-binding proteins to restore homeostasis. Mutations in their scaffolding lncRNAs (MALAT1 and NEAT1) may dysregulate some of these interactions, leading to cancer and
neurodegenerative diseases. In this project, we will elucidate the stress pathways dysregulated in cancer and neurodevelopmental diseases, the molecular mechanism behind specific RNA-protein interactions that cause disease, and develop tools to interfere with such interactions.
